A number of dietary factors influence immune function. Inflammatory mechanisms have been associated with the development of allergies and asthma. In vitro exposure of dendritic cells (DCs) to diet-derived molecules may be useful in predicting the effects of dietary triggers of inflammation in vivo. The objective of this study was to determine if macromolecules derived from a number of foods, common to Western diets, could alter the in vitro differentiation of DCs from peripheral blood. In addition, the effect of these foods on cytokine production by DCs was evaluated. We used a series of commercially available foods to explore potential relationships between foods and DC function. Dietary factors were selected for their known effects on DC function. DCs were differentiated in the presence of individual foods and cytokine production was assessed. Dose-response studies showed that foods affected DC function in a variety of ways. The ratio of omega-3 to omega-6 fatty acids in the diet was an important determinant of IL-10 production. The omega-3:omega-6 ratio had a significant effect on IL-12 production and on GM-CSF and IL-4 production from LPS-activated monocytes.
